DSPE-PEG-COOH is one of the PEG conjugates of reactive phospholipids that can react with primary amine groups. DSPE (1, 2-distearyl-Sn-glycerol-3-phosphate ethanolamine) is a highly hydrophobic 18-carbon phospholipid, on the other hand, with PEG connectors that provide good hydrophilicity and water solubility. Using EDC chemistry, the carboxyl group (-COOH) at the end of another PEG reacts with the primary amine group, so PEG DSPE can be easily incorporated into liposomes and other nanoparticles through the reaction of carboxylic acids and amines.
The carboxyl group at the end of PEG is activated by water soluble carbamyl diimide and can be attached to the amino group on the ligand (antibody).
DSPE Polyethylene glycol "liposomes" are commonly used to conjugate antibodies, proteins, peptides, or other substrates to the surfaces of liposomes and other lipid PEG nanoparticles. Pegylated phospholipids have longer blood circulation times and higher encapsulated molecular stability.
