Doravirine (MK-1439) is an HIV-1 single nucleotide reverse transcriptase inhibitor with IC50 of 12 nM, 9.7 nM, and 9.7 nM for wild-type reverse transcriptase and reverse transcriptase carrying mutations in K103N and Y181C, respectively. It is very specific and has very few off-target effects.

In Vitro: The IC50 of MK-1439 for DNA polymerase α, ß and γ in cells was greater than 100 μM. In the screening of protein targets (including enzymes, transporters, ion channels and receptors) in 110, the IC50 value of MK-1439 for most targets was greater than 10 μM, except for 5-HT2β, which was 2.5 μM for MK-1429 in the ligand binding assay. However, no 5-HT2ß activity was detected in the accumulation of inositol-1-phosphate in cells. In activated CD4+ T cells, peripheral blood monocytes, and macrophage proliferative transformation cell lines, MK-1439 showed no cytotoxicity at concentrations up to 100 μM.
