Calpeptin is a potent, cell-permeable calpain inhibitor against calsin I (porcine red blood cells), calsin II (porcine kidneys), papain, and calsin I (human platelets) with ID50 of 52 nM, 34 nM, 138 nM, and 40 nM, respectively. Calpeptin reduces apoptosis and intracellular inflammatory changes in muscle cells.

In Vivo: Calpeptin inhibits 20K phosphorylation in a dose-dependent manner in thrombin, iomycin, or collagen-stimulated platelets. In differentiated PC12 cells, Calpeptin promotes neurite elongation by inhibiting calpsin activity. In rat retinal ganglion cells, Calpeptin reduced apoptosis and maintained normal whole-cell membrane potential, thereby providing functional neuroprotection.
