Dacomitinib (PF299804, PF299) is a potent, irreversible panerbb inhibitor, most effective against EGFR with IC50 of 6 nM in cell-free trials. Dacomitinib inhibited ERBB2 and ERBB4 with corresponding IC50 values of 45.7 nM and 73.7 nM, respectively. Dacomitinib is highly effective on NSCLCs carrying EGFR or ERBB2 mutations (Gefitinib-resistant) and EGFR T790M mutations. Dacomitinib inhibited cell growth and induced apoptosis. Phase 2.
