Tacrolimus (FK506) is a 23-element macrolide that produces a new complex by binding to the immunosuppressant protein FKBP12 (FK506 binding protein), thereby reducing peptidyl prolyl isomerase activity in T cells. Tacrolimus also inhibits the phosphatase activity of calcineurin. Tacrolimus can induce autophagy of vascular endothelial cells.
