Unesbulin (PTC596) is a second-generation inhibitor of BMI-1, which can accelerate the degradation of BMI-1. PTC596 down-regulates MCL-1 and induces p53-dependent mitochondrial apoptosis. Mantle cell lymphoma cells were treated for 72 hours with an IC50 of 68-340 nM.
