Bempedoic acid (ETC-1002) is a small molecule that can be used orally to lower LDL-C and is an activator of AMPK in the liver. ETC-1002 effectively inhibited liver ATP-citrate lyase (IC50 = 29 uM).

In Vitro: ETC-1002 is a precursor drug in an inactive state that is converted into an ACL inhibitor in vivo by endogenous hepatic ACS activity (active state,ECT-1002-CoA).ETC-1002 inhibits thioglia-induced white blood cell migration into the peritoneal cavity of mice. ETC-1002 restored lipid AMPK activity, decreased JNK phosphorylation, and decreased macrophage-specific marker 4F/80 expression in a mouse model of diet-induced obesity.
